SUMMARY:

Phytate (Inositol hexaphosphate: C6H18O24P6 - IP6) is present in plants, especially in cereals such as soybeans, corn, wheat, oats and legumes. Currently, phytate is considered an antinutritional factor by binding to metals and minerals, such as iron, zinc, copper and especially phosphorus, reducing its absorption during digestion in monogastric animals. Excretion of undigested phytate can cause serious ecological problems due to excess phosphorus excreted into the environment. Phytases are enzymes capable of degrading phytic acid and are used in animal feed supplementation in order to improve digestibility through the release of nutrients such as phosphorus present in phytate form. Therefore, the use of phytases in poultry feed has been intensified in order to reduce supplementation with inorganic phosphors in feed and, consequently, the excretion of phosphorus. Therefore, a screening of phytases with relevant biotechnological characteristics aimed at a rational design of the proposal was carried out in this work. Phytases from filamentous fungi are currently promising for choice because they have desirable biochemical properties for application in the animal industry. Therefore, new clones of the yeast Komagataella pastoris GS115 containing the phytase gene of interest were used as a heterologous expression system, with the microorganism of origin the filamentous fungus Aspergillus nidulans. Several bioinformatic analyzes (in silico analyzes) of the gene sequence and protein were performed to optimize the mRNA sequences obtained in GenBank. The expression was performed with 0.5% methanol at different induction times. Preliminary tests of nitrophenylphosphate activity were performed.
