SUMMARY:

The growth in the use of pesticides and the like has been accompanying the great increase in the world population. However, the indiscriminate use of these substances can have major consequences for the environment and cause deleterious effects in non-target organisms. Fish, considered excellent bioindicators of environmental contamination, are widely used in toxicological tests due to the rapid response to low concentrations of contaminants, undergoing histological changes in key organs, such as gills, liver and kidneys. Some parameters, such as protein expression and quantification of certain cell types, are used as biomarkers, which indicate the level of damage suffered by exposure to pesticides. On the other hand, the development of new technologies for the treatment of contaminated water appears as a solution for the depollution of ecosystems. Thus, the objective of the present work is to analyze the effects of an abamectin-based formulation, a pesticide used in different cultures, on zebrafish (Danio rerio) and to test the effectiveness of copper ferrite in the treatment of residues from experimentation. For the development of the work, adult fish were obtained from accredited fish farming, acclimatized for a week with food twice a day. For the calculation of the LC50 (96 hs), the Probit method was used. Doses lower than the calculated LC50 will be used for the experiment where the concentration of abamectin in the aquariums will be obtained via high performance liquid chromatography (HPLC). After 15 days of exposure in a semi-static regime, the fish were euthanized to dissect the organs to be studied. The organs will be fixed, embedded in paraffin and sectioned. Cuts were stained with hematoxylin and eosin, submitted to the PAS technique and conducted for immunohistochemistry, for the identification of Glycoprotein-1 and HSP70. Organ samples will be lysed for protein extraction that will be quantified by Bradford for later identification of proteins of interest in the Western blot technique. The copper ferrite will be synthesized and characterized via a spectrophotometer. Chromatographic columns with sintered glass with variations in copper ferrite masses will be completed with samples from aquariums containing abamectin. The solutions that pass through the columns after adsorption will be read at the wavelength selected for the abamectin. The results obtained so far point to the strong toxic action of abamectin, causing behavioral and hepatic changes, as well as the expression of Pgp-1 and HSP70.
