TOXICOLOGICAL EVALUATION OF THE DELETORY EFFECTS OF THE BENZODIAZEPINES: CLONAZEPAM AND ALPRAZOLAM ON THE INITIAL DEVELOPMENT OF ZEBRAFISH (Danio rerio) - A morphological and biochemical study

Introduction: Pharmaceutical products have been observed in different environments, and environmental contamination by these emerging pollutants can occur in different ways, which consequently can cause serious ecological consequences to non-target organisms and even human health. Objectives: To evaluate the toxicological effects individually and of the cocktail of the benzodiazepines clonazepam (CLO) and alprazolam (ALP) during the initial development of zebrafish. Material and methods: To assess the toxicological effects of the drugs of interest on early embryonic development, lethal concentrations were calculated using the following concentrations: 0.125; 0.25; 0.50; 1.0 and 3.0 mg/L for ALP and 20; 50; 100; 120 and 150 mg/L for CLO. From CL 50, an embryotoxic assay will be performed for both benzodiazepines. Results: In embryos with 6-8 hours post fertilization (hpf) exposed to increasing concentrations of ALP and CLO in 96 h of exposure, it was possible to find a CL 50 of 2 mg/L for ALP and 95 mg/L for CLO . As main findings, we highlight the delay in hatching of embryos with ALP and CLO at the highest concentrations compared to the control group; as well as higher mortality in the first hours of the experiment; we also observed the presence of morphological alterations due to the contact of the embryos in the first stages of development with the drugs of interest, such as: column and tail deformation, loss of color indicating a reduction in pigmentation, edema of the pericardium and yolk sac. Conclusion: In this present work, it is possible to observe some adverse effects from exposure to these anxiolytic drugs during the initial development of zebrafish, making it possible to extrapolate to nature the damage caused to living beings due to exposure to these drugs released into the environment.
