EVALUATION OF BIOCHEMICAL CHANGES IN RENAL TISSUES AFTER THE DIGOXIN DERIVATIVE TREATMENT (BD-15)

SUMMARY:

Cardiotonic steroids (CTS) are compounds widely distributed in nature, being produced by
several types of plants and amphibians. Among the best-observed CTS, we highlight digoxin,
which is used for the treatment of heart failure and atrial fibrillation. Having proven the various
benefits of CTS, we cannot mention that they also affect cytotoxic effects. Therefore, studies
on compounds that reduce or eliminate such effects are relevant and growing, and the
synthesis of digoxin analogues has been considered a promising approach for the
development of more selective and less cytotoxic compounds. BD-15 is a new digoxin
derivative, which is being investigated for its neuroprotective effects and our work group has
already demonstrated protective effects preventing chemical ischemia in vitro. Thus, this work
aimed to evaluate possible biochemical changes in the kidneys of rats treated with the BD-15
molecule. Male Wistar rats (n=7) were divided into 2 CONTROL groups and treated with BD-
15 (100µl). On the third day the animals were submitted to the open field behavioral test and
after the third day the animals were euthanized and the kidneys were removed. A
homogenization process was carried out in a Potter-type tissue homogenizer, all the macerate
obtained was subjected to different centrifugation steps, resulting in membrane fractions and
homogenate used throughout the work. The first experiment was the verification of the total
protein levels in both the membrane fraction and the homogenate, the protein concentrations
of each sample were used to standardize the subsequent results. Subsequently, activity levels
of Na+, K+-ATPase, an essential enzyme for renal tissue, were evaluated. An increase in the
expression of enzyme activity was observed when treated with BD-15. The next experiments
were to evaluate the integrity of part of the renal enzymatic antioxidant system, through the
activity of catalase (CAT) and superoxide dismutase (SOD) and the content of reduced
glutathione (GSH) was also evaluated. The results found for these experiments were a
significant increase in CAT and GSH activity when treated with BD-15, however, SOD showed
no significant difference. Oxidative stress is a state of balance of pro-oxidant systems, to verify
the oxidative state of the renal tissue, analyzed the formation of hydrogen peroxide (H202) and
lipid lipoperoxidation levels (TBARS), in both did not obtain significant difference. The results
found suggest that BD-15 acts significantly with an increase in the expression of enzymes of
the antioxidant system. Overall, this study provides evidence for the action of BD-15 in relation
to such an important organ as the kidney.
