EFFECT OF THE USE OF ANTIOXIDANTS ON Na,K-ATPase ACTIVITY AND STRESS OXIDATIVE IN EXPERIMENTAL MODELS OF EPILEPSY AND PARKINSON'S DISEASE

SUMMARY:

Neurological disorders have a high prevalence and incidence, leading to important impacts on the individual's health and greatly increasing the mortality of this population. Among these disorders are epilepsy and Parkinson's disease. Epilepsy is a neurological disorder characterized by epileptic seizures due to neuronal
hyperexcitability, which may be related to changes in Na,K-ATPase activity and oxidative stress. We conducted a systematic review and meta-analysis to investigate the impact of exogenous antioxidant use on oxidative stress, Na,K-ATPase activity, seizure latency, mortality, and number of seizures in rodent models of epilepsy. In total,
561 references were identified. Considering the study design (PROSPERO/CRD42022356960), 20 articles were included for the Systematic Review and 12 studies for the meta-analysis using MEDLINE, EMBASE, Web of Science, Science Direct, and Scopus databases as sources, following PRISMA guidelines. The metaanalysis shows that in epileptic animals there is significant increase in nitric oxide and malondialdehyde levels, with reduction of GSH and Na,K-ATPase in all areas evaluated and reduction of superoxide dismutase (SOD) only in the whole brain. The use of antioxidants allowed significant reduction of nitric oxide and malondialdehyde,
and increase of GSH, SOD and Na,K-ATPase. In addition, there was a percentage decrease in the number of seizures and mortality, and meta-analysis showed longer latency to seizures in animals using antioxidant therapy, indicating that this treatment minimizes the biochemical and behavioral deleterious effects of epilepsy. In relation to Parkinson's disease, there is a loss of dopaminergic neurons in the substantia nigra, leading to a depletion of dopamine in the striatum, leading to motor dysfunction and cognitive deficits. The etiology may be associated with genetic factors, and among the genes compromised are those associated with Na,K-ATPase. A systematic review and meta-analysis are being conducted to evaluate the impact of protective and treatment
use of antioxidants on Na,K-ATPase activity and oxidative stress, and to assess behavioral changes. This study is registered (PROSPERO/CRD42022357180) and is PRISMA compliant. 109 references were identified, including 16 studies for the systematic review and 12 for the meta-analysis, using the same databases as above. The analyses are ongoing.
