SUMMARY:

Natural products have played an important role in human civilization since ancient
times. They are often used to prevent and treat diseases, even without proven safety
of use, prior knowledge of their mechanisms of action, potential toxicity of their
metabolites, and side effects. To this day, they play a key role in the development of
new drugs and pharmaceutical products, being promising molecules, especially for
discovering treatments for diseases that do not have many options available in the
market. Among natural products, some classes, such as flavonoids, stand out due to
their wide range of proven beneficial actions on the human body. Morin belongs to the
class of flavonoids and is a very promising compound, with high pharmacological
activity, having potent antioxidant action and low cytotoxicity. Therefore, the aim of this
work was to evaluate, in vitro, the genotoxicity, mutagenicity, antigenotoxicity, and
antimutagenicity of morin in non-tumoral Chinese hamster lung fibroblast (V79) and
human colon adenocarcinoma cell line (RKO-AS45-1). For this purpose, the cell
viability test with trypan blue was performed; alkaline comet assay and micronucleus
test with cytokinesis block, followed by evaluation of the cell division index. In a general
view of the results, morin showed genotoxicity only at the highest concentration in the
RKO-AS45-1 lineage and at all evaluated concentrations in the V79 lineage. When the
antigenotoxic effect was evaluated, there was a significant reduction in DNA damage
in both lineages. In the post-treatment, it was possible to observe the same profile of
damage reduction, but so far, only the V79 lineage has been employed. In the
micronucleus test, it was observed that the compound induced damage to the genetic
material of cells exposed to higher concentrations. Morin has shown to be a compound
with great potential and promising to be further studied and possibly used in the future
for the development of drug prototypes that could be used for different health problems,
due to its high biological activity.
