SUMMARY:

The use of medicinal plants for the treatment of illnesses is a long-standing one, but proof of their effects has been reported more recently. Thus, herbal medicine has great potential to improve the quality of life of human beings. Ageratum conyzoides is a plant of the Asteraceae family, popularly known as “menstrasto”, “erva de são joão” or “caatinga de bode”, used in many regions of Brazil in the treatment of numerous pathologies. The uterus, a constituent organ of the female reproductive system, is formed by tissue layers, one of which is muscular, the myometrium. Uterine contractility is important for the physiological processes that make up the menstrual cycle, being influenced by hormonal dynamics, but contractile dysfunctions are associated with gynecological pathologies. These pathologies compromise the spastic activity of the uterus, generating numerous discomforts, such as exacerbated menstrual cramps, which negatively influence the woman's well-being. Thus, the present study aims to study the effect of A. conyzoides essential oil on uterine smooth muscle and the influence of the estrous cycle stage on its action, from the induction of contraction by KCl (potassium chloride). Female Swiss mice from approximately 60 days of age will be used. According to the stage of the estrous cycle, the mice will be euthanized with isoflurane for later isolation of the uterine horns. The organ will be fixed and placed in an organ bath containing nutrient solution, where treatment with A. conyzoides essential oil and/or calcium channel blockers will occur before stimulation by KCl. In this way, the analysis of the effect of A. conyzoides essential oil on the contraction of the smooth muscles of the uterus will be carried out, according to the hormonal dynamics of the estrous cycle. Based on the knowledge obtained from this study, Ageratum conyzoides can be used to treat dysmenorrhea, in addition to other pathologies related to smooth muscle dysfunction of the female reproductive system.
